Studies on The Promotion of Transformation of Initiated Rat Liver Epithelial Cell by Phenobarbital, Butylated Hydroxyanisole and Butylated Hydroxytoluene

ABSTRACT
Newborne wistar rats were injected (i.p) with a single dose of 2-acetylaminofluorene (2-AAF) for in vivo model of initiation, one week later, the liver epithelial cells were obtained by a trypsinization of the pooled livers and used for in vitro model to evaluate the promotion of cell transformation by phenobarbital (PB), butylated hydroxyanisole (BHA) and butylated hydroxytoluene (BHT). In the absence of promoting treatment, the initiated liver epithelial cells remained untransformation at least during 20 weeks of culture. At the certain concentration, not only the growth and proliferative activity of the initiated liver epithelial cells were effected but also the marker of the transformation and malignant tumor such as the activity of r-glutamyltranspeptidase, epidermal growth factor binding and DNA increasing were induced in a dose-response manner by in vivo-in vitro combination model.
